Experimental System

One transcription factor under study is the glucocorticoid receptor. When activated by hormone, this receptor binds to the mouse mammary tumor virus (MMTV) promoter. To visualize this binding, we are using a cell line containing multiple repeats of the MMTV promoter, each driving a ras reporter gene (schematic above). This cell system was developed by Dr. Gordon Hager of the Hormone Action and Oncogenesis Section, and details can be found on his laboratory's site.
In live cells the array can be visualized as a distinct, bright structure (GFP-GR panel). These structures are the bona fide array because they produce array-specific transcripts, as detected by RNA FISH.
